Dorsal root ganglia are a complex and highly interconnected set of neuronal cells that are linked into peripheral nervous system pathways involved in both pain and the regulation of autonomic nervous system activity. In the present study, embryonic, as well as adult mouse dorsal root ganglia were explanted into organotypic culture. These explants were maintained for periods ranging from 3 days to 6 weeks, and were then immunohistochemically analyzed using antibodies directed against GAP-43, calcitonin gene-related peptide, and glial fibrillary acidic protein. Both embryonic and adult ganglia displayed high levels of calcitonin gene-related peptide-like immunoreactivity. A majority of adult ganglion cells also expressed GAP-43, whereas all embryonic-derived ganglia displayed GAP-43 immunoreactivity. Glial fibrillary acidic protein-positive cells were primarily located in the periphery of the ganglion. Adult ganglia displayed a greater number of GAP-43-positive cells than the embryonic ones. In addition, we could clearly distinguish five major distinct areas of the dorsal root ganglion: the cell body of the neurons, the peripheral and central glial limitans, and peripheral and central perineuronal nets.
